The Nephilim, by that name, are mentioned two or three times in the Bible. In Genesis 6, they are the demigods produced by interbreeding between divine beings and human women. In Numbers, they are the gigantic indigenous inhabitants of the land of Canaan. They may also appear in Ezekiel.

People often connect the word to the root n-p-l, "to fall", but I don't know that there's any actual evidence the word came about that way.

Ideally, I'd like to write more about the Nephilim once I've finished by working through chapters one by one in the Hebrew Bible. But what could I say that isn't already nicely covered in DDD?
